Samples from Scratch

I've been hearing a lot of artists saying they make their own samples. I know Madeon has said this before, just as an example. Does anybody have any sort of insight as to what they mean when they say something like this? Does creating your own mean using an existing sample and tweaking it? Using a plugin to generate the sound? Or some other magical means of generating a sample? Don't know, just thought I'd ask.
