Getting A Sound Back 'In Tune'

This question applies not just to GMS but to Sytrus or any other app. It's something that has puzzled me for ages...and no doubt a lot of others. It's the single biggest reason why I give up on some pieces.

OK...so lets say I've messed about with a sound. Made it sound a little different or better, or whatever. But, the new sound is clearly out of tune. Or, other way round....the original sound I was messing about with may have been out of tune and I want it in tune.

There seems to be no easy means of getting it back in tune. Often, detuned sounds still sound out of tune even with 'detune' turned off. Is there some simple technical means of retuning it...or do I have to go solely by ear ?

OH....and that brings me on to this song.....I cannot for the life of me reproduce the piano/guitar ( no idea what it actually is ) type sound let alone the actual notes. What are the 4 ( it may be more ) notes being played over and over ? Must be 'detuned' in some way.
